Novelty:

Transgenic mouse line expressing the eGFP reporter that is controlled by an NG2 promoter. This mouse model is a useful resource for in vivo as well as in vitro study of neuronal development and function.

 

Value Proposition:

Neuroglial cells like pericytes and oligodendrocytes express NG-2 proteoglycans in progenitor cells but the gene expression is down-regulated as the cells mature. Therefore, NG-2 expression marks the development and functionality of these neuronal cells. This invention is a transgenic mouse line expressing the eGFP reporter under the control of a NG-2 promoter marking cells that actively express NG-2 proteoglycans. Other advantages include:

• Easily detectable fluorescent reporter
• Facilitates in vivo and in vitro study of neuronal gene expression and dynamics
• eGFP expressing cells can be sorted for genetic analysis

 

Technical Details:

Johns Hopkins researchers have established a line of transgenic mice in which a membrane anchored form of eGFP is expressed under the control of the NG-2 promoter. EGFP is a mutant of GFP protein that provides increased fluorescence and photostability. In these mice, oligodendrocyte progenitors and some pericytes express eGFP and help in the study of these neuroglial cell development and dynamics.
