Technical Details:
DNA methyldtransferase1 (DNMT1) is a maintenance methyltransferase enzyme; additional roles for DNMT1 in gene expression are not understood clearly. DNMT1 is shown to have repressive activity, and deletion of its catalytic domain does not abolish this activity. In this study Johns Hopkins researchers examine the repressor function of catalytically inactive DNMT1 by generating this technology consisting of wtDNMT1 and catalytically inactive DNMT1 constructs with N-terminal FLAG tag inserts cloned into a pEF1á IRES-puro vector.
