Novelty:

This invention develops a method to measure the concentrations of various forms of Amyloid Precursor Proteins (APP) in cerebrospinal fluid (CSF).

Value Proposition:

Alzheimer's disease (AD) is the most common form of dementia. There is no cure for the disease. It worsens as it progresses, and eventually leads to death. APP plays an important role in AD formation. However, the concentration of the various forms of APP that occur in brain is not clear, nor if they can be differentiated in CSF. This invention develops a method to measure the concentrations of various forms of APP in CSF. Advantages of this technology include:

- Allowing differentiation between various forms of APP
- Facilitating the understanding of AD formation and pathogenesis
- Potential in providing therapeutic targets for AD

Technical Details:

The JHU inventors have developed a method to measure the concentrations of various forms of APP in CSF. APP is a single-pass transmembrane with three major isoforms. Sequential cleavages of APP protein by proteases generate specific soluble proteins and peptides which are used as biomarkers for AD. Control and dementia CSF samples were collected from the Johns Hopkins hospital. A multiplex, mass spectrometry-based method was developed to quantify the five different peptides from APP protein which allowed the differentiation between the various cleaved peptides. Conditions have been optimized to ensure data accuracy and reproducibility. Measurement of APP protein in CSF will help to unveil the mechanism of AD as well as other dementia related diseases.
